A fantastic new #zebrafish model of uveal #melanoma from the lab of @TheKennyLab. This demonstrates the power of modeling rare but difficult to treat cancers, opening up new avenues to understand the biology and find new treatments. It has been really nice seeing this story evolve and congrats on this beautiful work: aacrjournals.org/cancerres/a…

A special thanks to Board Chair, Doug Brodman and his wife, Terry, for hosting their fifth fundraiser in Naples, Florida. Since its inception, this event has raised more than $300,000 to support young melanoma investigators—fueling breakthroughs from the discovery of a promising immune cell linked to improved patient outcomes to the development of a clinical trial enhancing TIL therapy for advanced melanoma. Florian Karreth, PhD (@KarrethLab), MRF research grant recipient and researcher from @MoffittNews shared updates on the impact of basic science research and the profound impact that early funding of innovative science has on long term outcomes for melanoma patients.

Using this model, we identified two main UM cell states, melanocytic and neural crest-like, that may be associated with growth vs metastasis.

We also detected disseminated UM cells in the liver, making this a great model to study UM dormancy and metastasis

Live from #AACR25: Kaizhen Wang (@Kaizhen20) from the @KarrethLab discusses how MYBL2 drives melanoma through a surprising mechanism, non-canonical RNA binding. This newly uncovered function reveals how MYBL2 reshapes gene regulation and promotes tumor growth, offering fresh insight into melanoma progression. Key insights: •MYBL2 is pregulated and plays an oncogenic role in melanoma while represents a melanoma vulnerability • MYBL2 binds RNA in melanoma but not in melanocytes, and it promotes melanoma growth through its RNA binding function. • RNA binding of MYBL2 enhances its association with chromatin and drives a transcriptional program which associated with poor patient outcomes. • Distinct melanoma RNA background is the main determinant to enable MYBL2 RNA binding in melanoma cells Read more: bit.ly/4cSt6eP #MoffittAACR25

New preprint from the lab just posted to bioRxiv. Our first foray into antisense RNAs, led by postdoc Manon Chadourne, with implications for mRNA translation. Great collab with @ajaeger40 and his lab!
CDH3-AS1 antisense RNA enhances P-cadherin translation and acts as a tumor suppressor in melanoma biorxiv.org/cgi/content/shor… #biorxiv_molbio

Ribosome profiling confirmed the effects of CDH3-AS1 on P-cad translation but also uncovered a global effect of asRNAs: transcripts, especially those with longer 5'UTR, that have an asRNA overlapping with the 5'UTR, are translated more efficiently.

More work is needed, but asRNA appear to be key regulators of mRNA translation, potentially to deal with secondary structures in the mRNA 5'UTRs.
