Pyrithiamine-resistant transformants are selected on RM supplemented with pyrithiamine and then purified by streaking for single colonies on AMM supplemented with pyrithiamine. gDNA is then extracted from purified pyrithiamine-resistant transformants and those that have integrated the marker-free construct at the locus of interest are identified by PCR or other genotyping methods. A red circle indicates the expected product size for a co-transformant; on average, 11% of transformants are successfully co-transformed.
