When one of you posts the results of what you think needs to be checked, I'll pay attention but it appears neither of you have run this until you were called out for not having any familiarity with the tool. Yes.. You can select RevComp but its still ssDNA and too large as dsDNA

Here is another classic Gaff from Funk. Dr. Lee demonstrates Funk can't read an electropherogram. No Funk.. When you Sanger sequence from a forward read, you do not get a trace that include the forward primer. Only the revcomp of the reverse primer at the 3' end.

3/ look at the sequences - can you see the polyA tail? (stretches of Ts or As), can you identify the 5' and 3' cDNA primer? Here's an example from UHRR Iso-Seq data where you can see two reads with the polyA tail (as Ts), so we know these reads are revcomp of transcript

Always wanted to Polymerase methyl incorp one strand of DNA tethered to its native revcomp and run it through pores. Could do methyl A, Methyl C, Inosine and a variety of analogs that break up 2ndary structure and provide better error correction.
