A previously uncharacterized DNA element adjacent to enhancers, termed the "signpost", functions to guide transcription factor binding.

OSKM factors leverage motif orientation within chromatin loops to target pluripotent enhancers.
GETM factors use looping junctions to locate trophectoderm enhancers.

Happy to share our recent collaborative study with the Cedar and Stanger labs, identifying the barrier responsible for incomplete epigenetic reprogramming in both in vitro and in vivo transdifferentiation models.

Interestingly, the lack of demethylation, rather than de novo methylation, on key developmental regulatory elements, plays a crucial role in the incomplete reprogramming process. Kudos to all the authors for this surprising and important discovery!👏 #Epigenetics#Reprogramming
