New function for non-coding or "Junk" neutral DNA. A previously uncharacterized DNA element adjacent to enhancers, termed the "signpost", functions to guide transcription factor binding.

(3/n) Our study reveals that transcription factor combinations use distinct strategies to locate enhancers: OSKM factors leverage motif orientation within chromatin loops to target pluripotent enhancers. GETM factors use looping junctions to locate trophectoderm enhancers.
