Researchers optimized a method to isolate nuclei from flash‑frozen mouse colon tissue for single‑nuclei RNA sequencing using  Miltennyi’s lysis buffer and Octo-dissociator with chillers. The protocol improved nuclei RNA quality and counting prior to fixing nuclei for the 10x. Immune T cells can be genetically engineered to express “T cell receptors (TCRs)” that target specific molecules on cancer cells, but these cell-based therapies have shown limited clinical efficacy for patients with solid tumors, hindered by insufficient trafficking, persistence and anti-tumor function of the engineered T cells. @fredhutch investigators have now designed a chimeric TCR-containing engineering vehicle that improves tumor control in multiple laboratory models, supporting its broad utility.
