It's based on the emission spectra of the fluorescent dyes used in ABI gel sequencers - yellow was changed to black as it's hard to see a yellow trace on a white electropherogram (but not on a black gel background).

I'm having flashbacks to the FASTQ quality encoding conflict dx.doi.org/10.1093/nar/gkp11… - it seems @EarlhamInst's @brickopore didn't follow the LEGO block colour to DNA base convention established by @MonsterDNALab & @samstudio8, but invented their own! github.com/SAMTOMINDUSTRYS/m…

That is correct. I believe he used a 'string and bead' approach although the reader was a lego based system. I do wonder on any infringements over the method of pinning the beads to the string though... ;-)
