This week in @NatureProtocols we are sharing a detailed method for transplanting human stem cell–derived neural organoids into the somatosensory cortex of newborn rats as well as assays to functionally probe host circuit integration of the graft and test behavor. This is the 3rd article in a series to share step-by-step protocols for generating regionalized neural #organoids and #assembloids (links below), and is also the basis for our international Stanford @BrOrganogenesis hands-on course here on campus. After almost a decade, I have made the decision to leave OpenAI.  The company’s trajectory has been nothing short of miraculous, and I’m confident that OpenAI will build AGI that is both safe and beneficial under the leadership of @sama, @gdb, @miramurati and now, under the excellent research leadership of @merettm.  It was an honor and a privilege to have worked together, and I will miss everyone dearly.   So long, and thanks for everything. I am excited for what comes next — a project that is very personally meaningful to me about which I will share details in due time.

Today, I’m thrilled have our lab’s work featured on the cover of @Nature In this article, we have developed a therapeutic approach for a severe condition called Timothy syndrome. This has been a long journey. It started with making hiPS cell-derived neurons in 2D cultures 15 years ago when I was a postdoc and continued with later creating in the lab #organoid and then #assembloid models of this disease caused by a mutation in a calcium channel. Over time, these human cellular models and the biological insights we gained enable us to design a therapeutic strategy, which we validated in vivo following circuit integration of patient-derived organoids into the rat brain. Grateful for the incredible work led by @XiaoyuChenxy and @fikribirey in the lab! And the many close collaborators who participated to this line of research over the years.

There are now hundreds of genes associated with autism & neurodevelopmental disorders but what key processes in brain development they each disrupt remains unknown. In an article in @Nature today, we start tackling this question by combining forebrain #assembloids with CRISPR screens to map >400 disease genes onto stages of human cortical interneuron development. Using engineered lines, regionalized neural #organoids and over 1,000 #assembloids, we discovered a group of disease genes that interfere with GABAergic neuron generation or migration, including some surprising disease mechanisms.
