CONGRESS | #EHA2026 | POSTER Joshua Richter presents findings from a matching-adjusted indirect comparison of belantamab mafodotin plus bortezomib and dexamethasone (BVd; ESS = 47) vs teclistamab plus daratumumab (Tec-Dara; n = 291) for the treatment of RRMM. Following adjustment, BVd was associated with comparable OS, with RMST ratios of 1.04 at 1 year (p = 0.141), 1.04 at 2 years (p = 0.277), and 1.01 at 3 years (p = 0.801), and lower rates of treatment-emergent infections of any grade (81.3% vs 96.5%; p<0.001) vs Tec-Dara. These findings suggest BVd may offer a BCMA-directed treatment option for patients where infection risk is of particular concern. CONGRESS | #EHA2026 | POSTER Marina Kremyanskaya presents results from Arm 2 of the MANIFEST study, evaluating pelabresib added to ongoing ruxolitinib in suboptimal responders (N = 87). - Among 83 evaluable patients, 18.1% had SVR35 at Week 24 (16.4% TD; 21.4% non-TD); overall, 28.8% of patients had SVR35 at any time during treatment. - Median spleen response duration was 180.9 weeks. - At Week 24, 37.6% of evaluable patients (32/85) achieved TSS50 (36.2% TD; 40.7% non-TD). - Add-on pelabresib to ruxolitinib enabled 26.1% of TD patients to convert to TI. - The most common Grade ≥3 TEAEs were thrombocytopenia (39.1%), anemia (23.0%), and pneumonia (9.2%). Findings show that pelabresib added on to ongoing ruxolitinib demonstrated meaningful clinical benefit in a difficult-to-treat patient population, supporting further evaluation of pelabresib-containing combination approaches beyond JAK inhibition alone.
