We found that in addition to Fgf/Mek, Erk is activated by Aurora kinase A to promote differentiation. During cell division, Erk localizes at the centrosomes, where its activation depends on Plk1.

Pause and play: insights into mammalian embryonic diapause

Nutrient deprivation, suppression of the mammalian target of rapamycin complex 1 (mTORC1) or Myc activity can trigger dormancy response in mouse embryos. Diapausing embryos activate the leukemia inhibitory factor/signal transducer and activator of transcription 3/transcription factor CP2-like protein 1 (LifR/STAT3/Tfcp2l1) pathway and increase their dependency on fatty acid oxidation. During the first 3 days of diapause between E4.5 and estimated day of gestation (EDG) 7.5, the number of cells gradually increases and canonical Wnt signalling is temporarily activated. After EDG7.5, Wnt activity is downregulated, the epiblast undergoes epithelialisation and Yap translocates to the nucleus, governing the survival and reactivation capacity of the pluripotent lineage. The three background colours indicate dormant (grey), quiet (light yellow) and active (dark yellow) metabolic states.
