We purified ORF2p ‘core’ and determined of a 2.1 angstrom crystal structure, facilitating discovery of two novel domains we name the ‘tower’ and ‘wrist’. These flank the fingers-palm-thumb domains of reverse transcriptase (RT), which grip the nucleic acid like a “right hand”

L1 inserts a new, mutagenic copy of itself into the genome via ‘target primed reverse transcription’. Other outcomes contribute to pathology: failed insertions damage DNA, and we discovered here that aberrant cytosolic reverse transcription generates inflammatory RNA:DNA hybrids.
