Delighted to share our Cancer Cell paper published today, co-authored by Dr. Alexander Biederstädt @ABiederstadt_MD and Dr. Rafet Basar @rftbsr9 reporting the first genome-wide CRISPR screen in primary cord blood NK and CAR NK cells. We identify key genetic checkpoints including MED12, ARIH2 and CCNC and show that their deletion enhances CAR NK cell antitumor activity in multiple tumor models. Thanks @CellRepMethods for performing very professional review. Our work shows that cells transitioning into new transcriptional state in development or differentiation can be detected statistically via gene expression correlations in single cell rna-seq. Kudos @drmervedede etc

Excited to share our manuscript published in @ScienceTM, where we describe a novel NK immune evasion strategy in AML. We found that #BATF is a key transcription factor driving epigenetic reprogramming in NK cells in AML. This TF is induced by the canonical TGF-β/SMAD pathway and leads to persistent NK cell dysfunction. This dysfunction can be prevented by deleting BATF or TGFBR2 upstream, but once established, it cannot be reversed. Designed to overcome epigenetic scarring, we look forward to bringing this novel CAR-NK cell approach to the clinic to improve outcomes for AML patients. @mdandersonnews @lab_rezvani @bijender2050 #AML #NK #CRISPR #epigenetics #BATF
