crisprHAL ML model found poor activity at PAM sites with GATC sequences....seem familiar? GATC is DNA adenine methyltransferase site in bacteria. Large scale in vivo assays, Nanopore sequencing, and in vitro assays all show that G*ATC methylation impacts SaCas9 activity.

4/n Guess what? Size does matter - everything fits within a single AAV vector (TevSaCas9 repair guide). NO need for co-delivery of exogenous repair factors, accessory repair proteins/templates, or small/molecules/proteins to modify repair pathways.
