Beautiful work… This study is so exciting! Among many exciting findings, the authors found ultrastructural evidence for localization of TRiC/CCT in DA axons and the lack of axonal ribosomes in DA neurons, corroborating in vivo results from my graduate studies

Nat Biotech recently published a provocative study claiming single synapse RNA-seq. Unfortunately, there is no evidence demonstrating droplet-based sequencing of single synaptosomes. They are likely studying random pieces of cellular debris. Our reply: nature.com/articles/s41587-0…

Happy to share an approach to high-throughput, joint single-nucleus RNA/DNA-seq. Basically: prep nuclei for snATAC-seq badly on purpose and feed to the 10x Multiome kit for RNA/ATAC. Gives high-complexity RNA/DNA libraries instead. Hat-tip @olsen_timr: biorxiv.org/content/10.1101/…
