When words escape me, or my own feel woefully insufficient, I can always rely on Abigail Johnston (@AMJohnston1315) to express them with eloquence, grace, and heart.
We celebrated the life of our dear friend, Amy Beumer, yesterday, and will carry the grief of this loss for years to come. There is something bittersweet about sharing space with friends we’ve met through a disease no one would ever wish upon another human being, because we simply don’t know if, or when, we’ll see one another again, or how many more opportunities we’ll have to hug, laugh, and love on each other. ❤️🩹
What binds us together is also what drives us: raising our voices for a common cause, ending #metastaticbreastcancer, while hoping for more time and better quality of life until that day becomes reality, whether in our lifetime or in the lives of those who come after us.

Exciting data over the past year from trials of novel endocrine agents 📊
Oral SERDs continue to show efficacy in ESR1-mutant HR /HER2- MBC, and we now have two FDA-approved oral SERDs: elacestrant and imlunestrant ✅
Vepdegestrant, a novel PROTAC inhibitor, has also outperformed fulvestrant as 2L therapy in ESR1-mutant HR /HER2- MBC and is very well tolerated. If FDA approved, this would offer a monotherapy option after progression on 1L CDK4/6i ET.
Combination strategies have shown boosted efficacy (EMBER-3, evERA), though with increased toxicity ⚖️
The phase 3 SERENA-6 trial evaluated an early-switch strategy, substituting camizestrant for AI in 1L CDK4/6i-based therapy for patients with ESR1m detected in ctDNA without radiographic progression. Camizestrant CDK4/6i prolonged PFS by ~7 months vs AI CDK4/6i.
While universal adoption remains debated, these data add to growing evidence that ctDNA testing may enable early-switch strategies before progression 🧬

Results from @TheTBCRC 049 trial of tucatinib/cape/trastuzumab for HER2 LMD now published in @NatureCancer. Most patients had improved neurological deficits, PFS was 4.4 months and OS 10 months, with 41% of patients alive at 18 months. nature.com/articles/s43018-0…
