Altogether, an overly activated complement system may connect different hypotheses on Long Covid pathomechanisms. While the results require further validation in larger patient cohorts, they provide a basis for new diagnostic solutions and possibly targeted treatments. 9/

By measuring antibodies against >80’000 viral epitopes we found increased IgG against #herpesviruses in Long Covid patients, particularly against CMV and EBV. Anti-CMV was associated with markers of antibody-mediated complement activation. 8/

Using #MassSpec, we found increased markers of tissue injury, including red blood cells, platelets, and endothelial cells. #FlowCytometry paired with #scRNAseq showed increased monocyte-platelet aggregates in Long Covid patients. 6/

Functional assays revealed increased complement activation. Increased C5bC6 serum levels contrasted with the decrease of C7 complexes. This pattern suggested increased incorporation of terminal complement complexes in cell membranes. 5/

Experimental validation revealed that C7 complexes were decreased in the blood of Long Covid patients. Long Covid patients that had recovered before 6-month follow-up, showed normal levels. 4/

We measured >6’500 proteins in the blood of COVID-19 patients during initial infection and 6 months later, compared to healthy controls. 6 months after infection, one protein stood out: complement C7. 3/
