CONGRESS | #EHA2026 | PRESENTATION Johannes Schetelig presents findings from a randomized non-inferiority trial evaluating PTCy-100 vs ATLG-30 as #GvHD prophylaxis in allo-HSCT recipients from unrelated donors (N = 640). In the ITT population, the 2-year OS was 68% with PTCy-100 vs 75% with ATLG-30 (HR, 1.341; test for non-inferiority, p = 0.85; test for superiority, p = 0.051). The 2-year GRFS was 48% with PTCy-100 vs 40% with ATLG-30 (HR, 0.85; p = 0.16). The 2-year cumulative incidences of relapse were 26% and 31% (HR, 0.86), respectively. The 2-year NRM was 13% with PTCy-100 vs 7% with ATLG-30 (HR, 1.86; p = 0.02). Overall, confirmatory testing demonstrated that non-inferior survival with a 5% margin for PTCy-100 compared with ATLG-30 could not be shown. CONGRESS | #EBMT26 | PRESENTATION Florent Malard reports final results from the phase III, multicenter, open-label ARES trial, evaluating MaaT013 as a third-line treatment option in patients with treatment-refractory GI-#aGvHD after systemic steroids and ruxolitinib (N = 66). ARES met its primary endpoint, with a Day 28 GI-ORR of 62%, exceeding the pre-established threshold of 22% (p < 0.0001). Most patients had Day 28 GI-CR (38%) or GI-VGPR (20%), with 5% experiencing GI-PR. The all-organ ORR at Day 28 was 64%, and GI-ORR was maintained in 47% at Day 56 and 44% at Month 3. With a median follow-up of 344.5 days, the median OS was not reached; OS was 54% at Month 12, and was higher among Day 28 responders vs non-responders (68% vs 28%). CONGRESS | #ASH25 | POSTER Everett Meyer presents data from a subgroup analysis of the phase III Precision-T trial of Orca-T versus tacrolimus methotrexate (Tac/MTX) in patients with acute leukemia (N = 182). At 1 year, Orca-T demonstrated improved rates of chronic GvHD-free survival (78% vs 38%; p < 0.0001), OS (94% vs 83%; p = 0.045), GRFS (63% vs 31%; p < 0.0001), and NRM (13% vs 3.4%; p = 0.037) compared to Tac/MTX. Orca-T vs Tac/MTX showed improved clinical outcomes across patient subgroups with varied demographic and clinical features, including age, sex, donor type, and conditioning regimen.
