ALT iPSC-derived neurons were transduced with AAV to express a fluorescently labeled protein thought to localize primarily to synaptic vesicles. iNeurons were imaged every 24 hours across a 2-week growth factor-deprivation study. The top movie represents immediately pre-deprivation (day 0) The bottom movie represents 10 days post-deprivation. *Interesting note: re-introducing growth factor rescued the transport deficits within 48 hours*

ALT iPSC-derived organoids were virally transduced to express an RFP-tagged late endosomal protein called Rab7. Later, organoids were dissociated into single cells and plated on IBIDI chamber slides. The iPSC-derived neuron shown here was repeatedly recorded for 7 days after adhesion with a LEICA SP8 to assess transport dynamics during new neurite/axonal sprouting.
