1/7 Try our new app for optimizing transgenes for C. elegans expression (wormbuilder.org/transgenebui…). The app uses algorithms from collaborators Henrik Bringmann (TU Dresden) and Dan Dickinson (UT Austin) - or codons from highly expressed ubiquitous genes.

6/7 The optimized sequence is annotated in the app or in a GenBank file that can be opened in gene editors (for example @ApEplasmid)

7/7 The app was developed by postdoc Amhed Vargas (code, github.com/AmhedVargas/) and transgene expression tested by @sonia_elmouridi. We'll write up the details but let us know if you find any bugs or have feature suggestions (no promises!).

4/5 We integrated a > 5.4 Mb simple plasmid array and @FaisalMAlkhaldi assembled it from long-read Oxford Nanopore sequencing. Tandem arrays and truncated plasmids as first shown by Stinchcomb in 1985. First baby steps in trying to assemble larger synthetic genomic regions.
