John Mascarenhas, Icahn School of Medicine, presents findings from the phase I AJX-101 trial of AJ1-11095m a type II JAK2 inhibitor, in patients with myelofibrosis who have been failed by a type I JAK2 inhibitor (N = 23).
No DLTs or discontinuations due to TRAEs occurred. Dose reductions due to AEs occurred in 57% of patients. The most common Grade ≥3 TEAEs were anemia (52%), platelet count decreased (30%), ALT increased (4%), and cough (4%). The best SVR35 and SVR25 responses at any time were 70% and 83%, respectively. At a median follow-up of 8.1 months, 78% of patients remain on study therapy. At Week 24, 65% of patients achieved a TSS50 score. VAF reduction was observed in patients with JAK2, MPL, and CALR Type 1 and 2 mutations.
