Precision dosing of recombinant vaccinia vaccine via pillar-guided microneedle patch confers SARS-CoV-2 immunity
柱ガイド型マイクロニードルパッチによる組換えワクチニアワクチンの精密投与がSARS-CoV-2免疫を付与する
SARS-CoV-2のスパイク遺伝子を載せた組換えワクチニアウイルスワクチン（r-DIs-S）を、溶解型マイクロニードルパッチで届ける方法を開発した研究報告。
3Dプリントした柱がワクチンを針先に集中させ、1枚あたり約9.4±1.8×10^6 PFUを精密に搭載できた。
乾燥も速く、針先への搭載効率は16.5%（従来2.0%）に向上し、作製後も感染性は約50%保たれた。
マウスでは5分貼付するパッチ2枚と4週目の追加接種で、スパイク特異的IgGと中和抗体が強く誘導された。
致死量のSARS-CoV-2感染でも全例が生存し、肺のウイルス量と病変が減り、体重も保たれた。
Using 3D-printed pillars, a dissolving microneedle patch precisely loaded a recombinant vaccinia vaccine (r-DIs-S) onto needle tips, improving drying and preserving viral activity. In mice, brief patch application plus a booster triggered strong spike-specific antibodies and neutralization. Vaccinated animals survived lethal SARS-CoV-2 challenge with reduced lung virus and milder lung damage.

Eine neue Studie (Phase1/2)
in @Thelancet (doppelblind,randomisiert) zeigt dass die Anwendbarkeit easy, die Resorption gut und das Nebenwirkungs Profil extrem klein ist.
Wenn sich das bestätigt wäre das ein Riesenschritt mehr Kinder zur Impfung zu bringen.
Nebenbei wird das eine nervliche Entlastung für Eltern und Ärzte!
