The final version of our protein transposition manuscript is now published in @ScienceMagazine science.org/doi/10.1126/scie… We’ve added a few new things since our preprint /1
Ever wanted to swap out an entire internal region of a folded protein for functional biochemical studies? Check out our preprint that has been 5 years in the making, in which we establish a protein editing platform via protein transposition, enabling the insertion of functional cargo or PTMs into internal folded protein regions. #proteinengineering #proteinediting biorxiv.org/content/10.1101/…

In lab's #FIRSTPREPRINT, we present methods to measure nanometer organization around & between specific proteins in condensates. We uncover unexpected heterogeneity for a liquid-like phase with local meshwork spanning 10-50nm, stemming from ribosome biogenesis in the nucleolus.

I'm very excited to share the news of my promotion. Much of the credit goes to the trainees that drove the research forward, and the invaluable mentorship that I received along the way from Michael Hecht, @davidrliu, @KapoorLab_RU, my Princeton colleagues, and many others!
We are so very pleased to announce that @PrincetonChem’s Ralph Kleiner has been promoted to Associate Professor with tenure. Congratulations Ralph and the @KleinerLabRNA! Story here: bit.ly/4bJ4NhZ

Here is a photo of me with Crystal Dai after her PhD hooding ceremony this week. She was one of the first graduate students in the group and led the development of our activity-based probing platform for RNA modifying enzymes.

Ever wanted to incorporate ncAAs into your protein but found the efficiency to be too low? Check out our new preprint, where we turned a simple observation in protein translation to an effective quadruplet codon translation strategy!

Excited to share the latest manuscript from my lab and our first in @ACSCentSci . We used 5-BrUrd-modified tRNAs as activity-based probes to study the mechanism and substrate specificity of a human dihydrouridine synthase enzyme. Congrats to Jingwei and Nathan! @PrincetonChem

In the first, we use chemoproteomic and transcriptomic methods to study human DUS proteins and globally map tRNA D sites. We then show that DUS proteins control the levels of a subset of tRNAs and codon-specific translation in a cell type-specific manner.

In the second, we elucidate molecular principles underlying how hDUS2, a DUS protein associated with human cancers, installs a specific D residue in the tRNA D loop.
