🧵 NEW PAPER in @BloodJournal! Why do IDH1 mutations almost exclusively drive myeloid #leukemia, while IDH2 mutations occur in both myeloid AND lymphoid cancers? #AML #Epigenetics Despite sharing the same biochemical mechanism, they are NOT equivalent. Here's what we found 👇

Decades of research have revealed the intricacies of how dendritic cells (DCs) function, but fundamental questions remain about how this group of cells develops in humans and how this process goes awry in disease. A thread on our new work! (1/9) biorxiv.org/content/10.64898…

12/ To summarise: → IDH1 is highly expressed in myeloid progenitors (unlike IDH2) → Mutant IDH1 epigenetically silences Cebpe → neutrophil block → The block persists after IDH1 inhibition alone → IDH1i hypomethylating agent fully rescues the defect

Grateful to @JianXuLab for this perspective highlighting how oncogenes, developmental ontogeny, and cellular plasticity intersect in pediatric leukemia, alongside the JMML study by @mp_hartmann and @dblipka1. @BCD_AACR @CD_AACR aacrjournals.org/bloodcancer…
