Yes! Just contact whoever you bought the smartcard from and ask them to change the stations it's valid to/from with immediate effect.

Your (brilliant) calculator says I would save over £500 by doing this, but I still don't really understand how to. Is it possible with a Smart Card?

I don't see why not, but you'll either have to switch to a route where smartcards are available, or switch back to paper (no idea if that's possible).
