@shouval_lab @SnehaPramod @hagargir and I reflect on the work of the ✨ TME community in a new review re fibroblasts in cancer @NatureCancer. 2025 was my yr to 🩵 the distraction of writing (IYKYK), so thx to all whose discoveries made this so gratifying. nature.com/articles/s43018-0…

RevMed just doubled overall survival in pancreatic cancer by using CypA-targeting molecular glues to potently drug oncogenic KRAS-ON, long deemed untouchable. One funny thing is that probably the most fundamental insight for daraxonrasib sits in a PNAS paper that's been cited merely 84 times. This showed that molecular glues can coax an endogenous protein to wrap itself around utterly featureless surface. The other is a 2017 Cell Reports paper (just 68 citations) on how Sanglifehrin A can be used to repurpose CypA's surface. A lot of the game-changing stuff seems niche and unglamorous at first. Greg Verdine is having a chembio Annus Mirabilis for his 2025-2026 streak: FOG-001, Daraxonrasib. He provided much of the foundational conceptual work behind taking out both Beta-catenin and KRAS*. *of course many others contributed immensely, but let's give some credit where credit is due!

Thrilled to share our new @NatureGenet paper! We mapped human skin at single-cell spatial resolution and found that anatomy is encoded in cell states and neighborhoods, led by PhD student @paularstrpo. Link: nature.com/articles/s41588-0… 1/9

Great work in Mol Cell from John Maciejowski's group delineating BAF dependent regulation of TREX1 and cGAS in micronuclei. Very important implications for immune responses to chromosome instability. cell.com/molecular-cell/abst…

🎉 Congratulations to MSK physician-scientist @KarunaMDPhD and colleagues, who are part of an international team awarded a @CancerGrand grant — a prestigious global award providing up to $25M over five years to tackle some of the toughest challenges in #cancerresearch. The team will explore new ways to “rewire” cancer cells and prevent treatment resistance in #colorectalcancer, one of the leading causes of cancer-related deaths. Learn more: bit.ly/4d1GX4p

New preprint on technologies to scale up CRISPR screens. We use them to map 665,856 pairwise genetic perturbations and outline a path to comprehensive interaction mapping in human cells. We also introduce an approach for cloning lentiviral libraries with billions of elements.

1/7 New York's biomedical research enterprise is at an inflection point. Federal funding uncertainty has hit NY harder than any other state. Grant reviews are at 30% of prior levels. PhD programs cutting admissions 30-100%. A bold $6B proposal aims to change that. 🧵
