(5/7): In cancer, mTOR functions as a proto-oncogene, and its aberrant activation promotes tumorigenesis. Activating mTOR mutations have been identified in multiple malignancies, including clear cell renal cell carcinoma, endometrial carcinoma, anaplastic thyroid cancer, colorectal adenocarcinoma, and lung adenocarcinoma. These mutations occur across multiple domains and alter kinase activity directly or indirectly.
