Ahead of @ASCO 2026, we asked leading AI models to forecast outcomes for major GI cancer trials and compared those predictions with the data presented at ASCO 2026. Overall accuracy: ➡️ Median survival: R² = 0.96 (n=6) ➡️ Hazard ratios: R² = 0.82 (n=3) Dataset: ➡️ 3 major GI cancer trials ➡️ 16 predictions ➡️ 3 AI-modeled KM curves ➡️ Claude served as the final judge Where AI got it right: EMERALD-3 (PFS, HCC) ▶️ AI: HR 0.65 ▶️ Actual: HR 0.70 EMERALD-3 (OS, HCC) ▶️ AI: HR 0.77 ▶️ Actual: HR 0.84 (NS) NCT06008119 (ORR, BRAF-mutant CRC) ▶️ AI: ~39% ▶️ Actual: 37.1% Where AI underestimated benefit: NCT06008119 (PFS, BRAF-mutant CRC) ▶️ AI: HR 0.62 ▶️ Actual: HR 0.37 This was the largest prediction gap in the GI dataset. Key takeaways: ✅ AI correctly identified the direction of benefit across the major GI readouts ✅ Accuracy was strongest for response-based endpoints and broad efficacy signals ✅ The largest miss came in BRAF-mutant colorectal cancer, where AI predicted success but significantly underestimated the magnitude of benefit ✅ Unlike some other disease areas, GI produced few major directional misses; most discrepancies involved the size of the treatment effect AI continues to show value in anticipating broad clinical outcomes, but estimating the magnitude of benefit remains challenging when therapies outperform historical expectations.
